Biwi No 1 is a story of Prem (Salman Khan) who is married to Pooja (Karisma Kapoor) and has two kids. He is the owner of one of the biggest model agency. He comes in contact with Rupali (Sushmita Sen), who is an aspiring model and joins Prem's agency. Eventually, Prem and Rupali fall in love. The main reason why Prem falls for Rupali is because she is a modern women and has a fashionable dressing sense. On the night of Karva Chauth, Pooja finds Prem at Rupali's house and asks him to choose between Rupali and his family. Prem chooses Rupali over his family. On the other hand, Pooja, with the help of Prem's best friend, Lakhan (Anil Kapoor), becomes a model and plots a plan to get Prem on the right track. She sends her mother-in-law and kids at Rupali's house. But the question remains will Prem realize his mistake? Will he reunite with Pooja or continue to stay with Rupali?
